Anti-proliferative and Apoptotic Effects of Valproic Acid on HeLa Cells

Background: Valproic acid (VPA), a branched short-chain fatty and histone deacetylase (HDAC) inhibitor, has diverse biological activities in human cells, including anti-cancer properties. Objectives: In the present study, we tested cytotoxicity of VPA on proliferation, cell cycle, apoptosis cervical cancer line, HeLa. Methods: HeLa line was cultured Dulbecco’s modified eagle medium (DMEM) effect (at 0 - 100 mM) evaluated, using 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay for 3 incubation times (24, 48, 72 h). The effects cycle arrest were flow cytometry. addition, alterations expression Bax, Bcl-2, p53, p21 assessed with real‐time polymerase chain reaction (PCR). Results: reduced viability cells concentration- time-dependent manner, IC50 values at 24, h 32.06, 21.29, 14.51 mM, respectively. Further, treatment remarkably increased arrested sub-G1 phase significant reduction G2-M populations. real-time PCR results demonstrated increase pro-apoptotic genes, p21, as well levels anti-apoptotic gene, Bcl-2. Conclusions: inhibits proliferation through induction intrinsic pathway p35-dependent manner.
